If there is no sales tax on clothing that is sold in new York state and is under $55 per piece. Do I need to collect sales tax on shirts that I am selling that I printed?
Say a business comes to Me and has me print 100 shirts. Should I collect tax? What about an individual.

Wouldn't it depend on how much you're selling your shirts for? If what you say is correct about no tax for less than $55 per piece, why are you asking?
