What would be the best way to do this? I believe it was created in photoshop and i can get the original file so I have the best possible graphic to start. But I am having a hard time wrapping my head around how to go from pc to screens to make this look right.

Any help would be much appreciated.

Separation Studio would be my choice of colour separation program. This image would be easy to colour sep into 4 colours, underbase white, red, yellow, top white. All tones output to 55lpi - 22.5º on 230 mesh screens.
