I was just wondering....

Obviously it costs more to have your five colour design screen printed, than it would a one colour design.

Let's say you have a variety of shirts in your shop - some one colour designs, two coloured designs, 5 colour...etc..etc..

Do you make the 4 and 5 colour ones more expensive than the others? Or do you just charge a flat rate for all of your shirts?

What's your approach regarding this and what do you think is better?

I would agree with joe. although you could set a scale based upon intricacy of the shirts. so your basic 1,2,3 color design shirts all have the same prices. the shirts with a bit more colors maybe add a buck or two, something like that.
