Default Printing 2 or 3 colors & transparencies

Please Help ! I usually only print simple 1 color designs. I want to do 2 and 3 color designs. I don't know anything about separating the colors for different transparancies. I'm not looking for process color separations. How do I take a logo using photoshop, or adobe illustrator 9.0 and make the different separate transparancies with out the colors overlapping. In laymans terms. Please

magic wand tool, select an area --> layer via cut. To select more than one area, hold the shift key and use the magic wand. For a black trap layer you want to add a 2-3px stroke.
