Vinyl works great for simpler designs or short run jobs. You can cut a transfer, weed it and press it in just a few minutes. It's very durable and looks good. Plastisol will take a lot longer to make the screens, print, cure, transfer, etc.
The cost benefit starts to swing towards plastisol somewhere around a dozen or two transfers. Vinyl can be VERY labor intensive and there's no savings in volume.

Yes. Vinyl is not the right choice for this application. A 5 color (possibly 6 if you are using adhesive) will not be cheap. Probably $6-$7 (USD) each. You might want to look at the freedom transfers that are being discussed right now.
It also might be cheaper to just do a regular screen print run if you know that you absolutely want 50 shirts printed up. The only advantage in going with transfers is that it would give you the ability to press on demand and not tie capital up in inventory.
